Decoding complexity in biomolecular recognition of DNA i-motifs with microarrays
The three-dimensional structure of the genome has a major impact on gene expression.
Predominantly, the human genome consists of duplex B-form DNA though it can also adopt several non-B conformations including DNA intercalated-motif (i-Motif). i-Motifs are formed by C-rich DNA sequences which are dynamic and pH-sensitive. They have been implicated in various cellular processes and are recognized by proteins and small molecules, impacting gene expression and potentially serving as therapeutic targets. The research using DNA microarray with 10 976 distinct i-Motif-forming sequences found in human genome and InnoScan 1100 fluorescence scanner, has provided insights into the binding preferences of i-motif-binding proteins and small molecules, enhancing our understanding of their biological significance.
